Nepafenac (0.1% w/v)
Active composition
Nepafenac is converted to its active form in the eye, where it inhibits cyclooxygenase enzymes, reducing the synthesis of inflammatory mediators.
Medical applications
Xinep Eye Drop contains Nepafenac 0.1% w/v, a nonsteroidal anti-inflammatory drug (NSAID) indicated for the management of ocular inflammation and pain associated with cataract surgery. Nepafenac is a prodrug that penetrates the cornea and is converted by ocular tissue hydrolases to amfenac, an active inhibitor of cyclooxygenase enzymes COX-1 and COX-2. By inhibiting prostaglandin synthesis, Xinep Eye Drop effectively reduces inflammation, pain, and photophobia following ophthalmic procedures. It is typically administered as one drop in the affected eye(s) three times daily, beginning one day prior to surgery and continuing for up to two weeks postoperatively, or as directed by a healthcare professional. Xinep Eye Drop is contraindicated in patients with hypersensitivity to nepafenac or any component of the formulation. Caution is advised in patients with known bleeding disorders or those using concomitant anticoagulant therapy due to potential increased risk of bleeding. Common adverse effects may include transient burning or stinging upon instillation, blurred vision, and eye irritation. Patients should avoid contact lens use during treatment unless otherwise instructed. Proper administration technique and adherence to prescribed dosing are essential to optimize therapeutic outcomes and minimize adverse effects.
