Gluconorm PG 1 Tablet PR is a combination oral hypoglycemic agent containing Glimepiride 1mg, Metformin 500mg, and Pioglitazone 15mg, indicated for the management of type 2 diabetes mellitus. This formulation leverages the complementary mechanisms of action of its components to achieve optimal glycemic control. Glimepiride, a sulfonylurea, stimulates pancreatic beta-cell insulin secretion, thereby reducing blood glucose levels. Metformin, a biguanide, primarily decreases hepatic glucose production and improves peripheral insulin sensitivity, contributing to enhanced glucose uptake by muscle tissues. Pioglitazone, a thiazolidinedione, acts as an insulin sensitizer by activating peroxisome proliferator-activated receptor gamma (PPAR-γ), improving insulin sensitivity in adipose tissue, muscle, and the liver. The combination is particularly beneficial for patients inadequately controlled on monotherapy or dual therapy, offering improved efficacy through multiple pathways. Gluconorm PG 1 Tablet PR should be administered with meals to minimize gastrointestinal side effects associated with Metformin.
