Metformin, a biguanide, primarily acts by decreasing hepatic glucose production, enhancing insulin sensitivity, and improving peripheral glucose uptake and utilization.

Teneligliptin, a dipeptidyl peptidase-4 (DPP-4) inhibitor, increases endogenous incretin levels, thereby promoting glucose-dependent insulin secretion and suppressing glucagon release, which collectively contribute to lowering blood glucose levels. The sustained-release formulation of Metformin ensures prolonged therapeutic effect with reduced gastrointestinal side effects commonly associated with immediate-release forms.
Glipti Next M 1000 Tablet SR is indicated as an adjunct to diet and exercise to improve glycemic control in adults inadequately controlled on Metformin alone or in combination with other antidiabetic agents. It is essential to monitor renal function prior to and during therapy, as Metformin is contraindicated in patients with significant renal impairment. Common adverse effects may include gastrointestinal disturbances, hypoglycemia (especially when combined with other antidiabetics), and rare cases of lactic acidosis.
This combination therapy offers a complementary mechanism of action, targeting multiple pathophysiological defects in type 2 diabetes, thereby optimizing glycemic management.
