Glimepiride, a sulfonylurea, stimulates pancreatic beta cells to increase insulin secretion, thereby reducing blood glucose levels. Metformin, a biguanide, primarily acts by decreasing hepatic glucose production and improving peripheral insulin sensitivity, contributing to enhanced glycemic control. Pioglitazone, a thiazolidinedione, further improves insulin sensitivity in adipose tissue, muscle, and the liver by activating peroxisome proliferator-activated receptor gamma (PPAR-γ).
