This formulation synergistically improves glycemic control through complementary mechanisms: Glimepiride, a sulfonylurea, stimulates pancreatic beta-cell insulin secretion; Metformin, a biguanide, decreases hepatic glucose production and enhances peripheral insulin sensitivity; Pioglitazone, a thiazolidinedione, improves insulin sensitivity in adipose tissue, muscle, and the liver by activating peroxisome proliferator-activated receptor gamma (PPAR-γ).
