Glibenclamide, a sulfonylurea, stimulates pancreatic beta cells to enhance insulin secretion. Metformin, a biguanide, primarily reduces hepatic glucose production and improves peripheral insulin sensitivity, thereby lowering basal and postprandial blood glucose levels. Pioglitazone, a thiazolidinedione, acts as an insulin sensitizer by activating peroxisome proliferator-activated receptor gamma (PPAR-γ), improving glucose uptake in adipose tissue and muscle.
