Gabapentin, an anticonvulsant agent, modulates neuronal excitability by binding to the alpha-2-delta subunit of voltage-gated calcium channels, thereby reducing neurotransmitter release and alleviating neuropathic pain symptoms. Nortriptyline, a tricyclic antidepressant, exerts analgesic effects through inhibition of norepinephrine and serotonin reuptake, enhancing descending inhibitory pain pathways.
