Ilevro Ophthalmic Suspension contains Nepafenac 0.3% w/v, a nonsteroidal anti-inflammatory drug (NSAID) indicated for the treatment of ocular inflammation and pain associated with cataract surgery. Nepafenac is a prodrug that is converted by ocular tissue hydrolases to amfenac, an active metabolite that inhibits cyclooxygenase enzymes COX-1 and COX-2, thereby reducing prostaglandin synthesis which mediates inflammation and pain. Ilevro is administered as a topical ophthalmic suspension, typically dosed once daily beginning one day prior to cataract surgery, continued on the day of surgery, and for up to two weeks postoperatively. This dosing regimen facilitates effective control of postoperative inflammation and reduces the risk of ocular pain. The formulation’s 0.3% concentration allows for enhanced penetration and sustained therapeutic levels within ocular tissues.
