Glimepiride, a sulfonylurea, stimulates pancreatic beta-cell insulin secretion, thereby reducing blood glucose levels. Metformin, a biguanide, primarily decreases hepatic glucose production and improves peripheral insulin sensitivity, contributing to enhanced glucose uptake. Pioglitazone, a thiazolidinedione, acts as an insulin sensitizer by activating peroxisome proliferator-activated receptor gamma (PPAR-γ), improving insulin sensitivity in adipose tissue, muscle, and the liver.
